The Mouse Biology Program (MBP) provides germline transmission testing for the chimeras produced from blastocyst injection and breeding of transgenic founders produced by microinjection.
Germline transmission testing
Two or three of the highest percentage (preferably >50%) 6 week old male chimeras are set with inbred females (strain will depend on stem cell lineage) in breeding trios. Breeding will continue until at least one heterozygous (founder) pup, or forty wild type pups are produced. Determining germline transmission can take as long as 120 days, and depends on the fertility and fecundity of the males. The customer will receive heterozygous pups and wild type controls which are ready to ship approximately 3 weeks after weaning.
Rate for germline transmission testing
For University of California investigators, we generally estimate around $900 per chimera tested, or $1300 for all other academic investigators.
